DMAN: a Java tool for analysis of multi-well differential scanning fluorimetry experiments
نویسندگان
چکیده
SUMMARY Differential scanning fluorimetry (DSF) is a rapid technique that can be used in structural biology to study protein-ligand interactions. We have developed DMAN, a novel tool to analyse multi-well plate data obtained in DSF experiments. DMAN is easy to install and provides a user-friendly interface. Multi-well plate layouts can be designed by the user and experimental data can be annotated and analysed by DMAN according to the specified plate layout. Statistical tests for significance are performed automatically, and graphical tools are also provided to assist in data analysis. The modular concept of this software will allow easy development of other multi-well plate analysis applications in the future. AVAILABILITY AND IMPLEMENTATION DMAN is implemented in Java to provide a cross-platform compatibility. It is freely available to academic users at http://www.structuralchemistry.org/pcsb/. To download DMAN, users will be asked for their name, institution and email address. A manual can also be downloaded from this site. CONTACT [email protected]; [email protected].

Sensitive Detection of Immunoglobulin G Stability Using in Real-Time Isothermal Differential Scanning Fluorimetry: Determinants of Protein Stability for Antibody-Based Therapeutics
Protein instability is a major obstacle in the production and delivery of monoclonal antibody-based therapies for cancer. This study presents real-time isothermal differential scanning fluorimetry as an emerging method to evaluate the stability of human immunoglobulin G protein with high sensitivity. The stability of polyclonal human immunoglobulin G against urea-induced denaturation was assess...
متن کاملDifferential Scanning Fluorimetry provides high throughput data on silk protein transitions.
Here we present a set of measurements using Differential Scanning Fluorimetry (DSF) as an inexpensive, high throughput screening method to investigate the folding of silk protein molecules as they abandon their first native melt conformation, dehydrate and denature into their final solid filament conformation. Our first data and analyses comparing silks from spiders, mulberry and wild silkworms...
متن کاملDetermination of Protein-ligand Interactions Using Differential Scanning Fluorimetry
A wide range of methods are currently available for determining the dissociation constant between a protein and interacting small molecules. However, most of these require access to specialist equipment, and often require a degree of expertise to effectively establish reliable experiments and analyze data. Differential scanning fluorimetry (DSF) is being increasingly used as a robust method for...
متن کاملElectrochemical Analysis of Tryptophan using a Nanostructuring Electrode with Multi-walled Carbon Nanotubes and Cetyltrimethylammonium bromide Nanocomposite
Multi-walled carbon nanotubes (MWCNTs) were immobilized on the surface of a glassy carbon electrode (GCE) in the presence of cetyltrimethylammonium bromide (CTAB) to form a MWCNTs-CTAB nanocomposite-modified electrode. The electrocatalytic response of the modified electrode towards tryptophan (Trp) was investigated by cyclic voltammetry (CV) and differential pulse voltammetry (DPV). The surface...
متن کاملAnalysis of Milling Process Parameters and their Influence on Glass Fiber Reinforced Polymer Composites (RESEARCH NOTE)
Milling of fiber reinforced polymer composites is of great importance for integrated composites with other mating parts. Improper selection of cutting process parameters, excessive cutting forces and other machining conditions would result in rejection of components. Therefore, machining conditions are optimized to reduce the machining forces and damages. This work reports practical experiments...
